New Discovery of Sterile Mechanism

Ten to fifteen percent married couples of childbearing age in the world are now obsessed by sterility, abnormal germ cell forming being one of the important causes. A scientific team led by Prof. Han Min and Prof. Xu Ren’er of Fudan Institute of Development Biology and Molecular Medicine has recently made breaking progress in the research of germ cell forming mechanism by applying genetics methods to the sterility study. This result has been published on Development Cell of June 4, with the title of SUN1 Is Required for Telomere Attachment to Nuclear Envelope and Gametogenesis in Mice, and is expected to be selected as “the noteworthy paper” by the upcoming issue of Science.

Fudan Hospital Faculty Publishes on Clinical Oncology on Liver Cancer

Prof. Fan Jia, Prof. Qiu Shuangjian and their doctoral student Gao Qiang from Fduan Liver Cancer Institute published a paper on the immune micro-environment and the prognosis of liver cancer on Journal of Clinical Oncology, marking a breakthrough in this research field in China, with the title of Intratumoral Balance of Regulatory and Cytotoxic T Cells Is Associated With Prognosis of Hepatocellular Carcinoma After Resection.

Journal of Clinical Oncology is the official publication of American Society of Clinical Oncology (ASCO) introducing the latest research findings in Clinical Oncology internationally. This paper is based on rich clinical practice and employing advanced tissue chip technology. In the past, research showed that there is a possible increasing of repressive T-lymphocyte-regulatory T-lymphocyte. Such inhibition of T lymphocytes greatly reduces the presence of the anti-tumor immune response, but there is not enough evidence and the clinical significance is not clear.

This research is aimed to investigate the prognostic value of tumor-infiltrating lymphocytes (TILs), especially regulatory T cells (Tregs), in hepatocellular carcinoma (HCC) patients after resection. In the paper, CD3+, CD4+, CD8+, Foxp3-positive, and granzyme B-positive TILs were assessed by immunohistochemistry in tissue microarrays containing HCC from 302 patients. Prognostic effects of low- or high-density TIL subsets were evaluated by Cox regression and Kaplan-Meier analysis using median values as cutoff.

According to the research results, CD3+, CD4+, CD8+ TILs were associated with neither overall survival (OS) nor disease-free survival (DFS). The presence of low intratumoral Tregs in combination with high intratumoral activated CD8+ cytotoxic cells (CTLs), a balance toward CTLs, was an independent prognostic factor for both improved DFS (P = .001) and OS (P < .0001). Five-year OS and DFS rates were only 24.1% and 19.8% for the group with intratumoral high Tregs and low activated CTLs, compared with 64.0% and 59.4% for the group with intratumoral low Tregs and high activated CTLs, respectively. Either intratumoral Tregs alone (P = .001) or intratumoral activated CTLs (P = .001) alone is also an independent predictor for OS. In addition, high Tregs density was associated with both absence of tumor encapsulation (P = .032) and presence of tumor vascular invasion (P = .031).

Hence the paper concludes that tregs are associated with HCC invasiveness, and intratumoral balance of regulatory and cytotoxic T cells is a promising independent predictor for recurrence and survival in HCC. A combination of depletion of Tregs and concomitant stimulation of effector T cells may be an effective immunotherapy to reduce recurrence and prolong survival after surgery.

This research finding arouses great attention and interest of the field. The editor’s note specially written for the paper pointed out that the paper further clarifies the core procedure and mechanism of cancer immune response. It is a guide to choose correct immune treatment after surgery for the high-risk patients with cancer recurrence.

The group on micro-environment and liver cancer metastasis and recurrence led by Prof. Fan Jia has been committed to the research on the interaction of host and tumor since 2003 as one of the first groups of this nature. The publication of this paper marks an important achievement for the present stage of research. Now a clinical random comparative research and a series of basic research based on the finding are being carried on.

“Rural Waste into Clean Energy” Project Launched

The Urban Environmental Management Research Center at Fudan Department of Environmental Science launched a topical study on "The Research and Application of the Transformation of Rural Waste into Clean Energy", which was funded by Shanghai Sci-Tech Commission with one million RMB.

The project aims at the research and development of an integrated series of methane fermentation equipment that applies to different natural villages. By the methane fermentation, rural waste and domestic sewage will be comprehensively transformed into rural energy, thus to provide clean and sustainable green energy and to provide long-term rural pollution control as well as an access to environmental resources and dual income.

Relevant experts pointed out that China is rich in biomass resources. Through the development of standardization and mass production of methane fermentation equipments, China’s gas industry can be scientifically developed and grown, which is also in line with China’s situation to promote renewable energy. The project has formally started and will begin its demonstrative program in Xiexi Village, Gangxi County on Chongming Island.
